When you turn it on only the colored part of the layer can be colored
Like uh if you paint a tiny dot and then turn transparency lock on and paint it with a bigger brush only the dot will be recolored
If that makes sense

In the layers tab on the right there's an opacity slider and two buttons and the lower button lets you duplicate the layers
